SIM media is a differential media that is inoculated by a stab needle and tests the motility of an organism as well as its ability to reduce sulfur and, with the addition of Kovac’s reagent (sold separately, item #871389), the ability to produce indole. The reduction of sulfur to hydrogen sulfide produces a black color. Organisms that convert tryptophan to indole produce a red color in the presence of Kovac’s reagent, and organisms that are motile have a growth pattern that radiates from the stab mark and cause the media to appear turbid. We recommend Salmonella typhimurium to demonstrate positive results for motility and sulfur reduction. Escherichia coli is positive for motility and indole production. Streptococcus pyogenes is negative for all 3 tests. It is recommended that prepared media tubes be used within 3 months of receipt for best results. Note: The species listed are not an inclusive list and are provided to assist novice users in selecting compatible media and bacteria.
